Abstract
There is increasing evidence to suggest that activin can function as an autocrine/paracrine regulator in various tissues, including the reproductive system. At the cellular level, activin acts via a family of activin receptor (ActR) subtypes which includes two type I (ActRI and ActRLB) and two type II (II and ILB) receptors. The role of activin in the human placenta is not clearly understood. In this study, the detection of inlubin/activin subunit and ActRI mRNA were examined in first trimester cytotrophoblasts, term cytotrophoblasts, extravillous trophoblast (EVT) cells, immortalized extravillous trophoblast (LEVT) cells, JEG-3 cells, decidual tissue, and decidual cells.

These studies examined the effects of prostaglandin-F[sub 2a] (PGF[sub 2a]) on progesterone and 178-estradiol (estradiol) production, as well as DNA and PGF[sub 2a]-receptor (PGF[sub 2a]-R) mRNA levels, in the human granulosa-luteal cell (GLC). Additionally, the interactions of PGF[sub 2a] with human chorionic gonadotrophin (hCG), gonadotrophin-releasing hormone (GnRH) and prostaglandin E₂ (PGE₂) were examined, with respect to progesterone and estradiol production.

Abstract
It is well established that LH action is mediated primarily by adenylate cyclase/cAMP. Conversely, the role of inositol phosphate/calcium in LH signalling has only recently been investigated. We examined the effects of gonadotrophins on intracellular calcium mobilisation in HEK293 cells transiently transfected with human wild type or chimeric gonadotrophs receptors (n=3400). Intracellular free calcium concentration was measured using fura-2 microspectrofluorimetric techniques. Human LH (2-4 μg/ml) arid CG (10 IU/ml) consistently evoked oscillatory calcium signals in HEK293 cells transfected with hLHr, whereas hFSH (2-4 μg/ml) failed to elicit any calcium responses.

Abstract
It is increasingly apparent that GnRH-II acts as an autocrine/paracrine regulator in non-pituitary tissues, in addition to its role in the regulation of gonadotropin synthesis, and is an important player in cancer cell biology. High levels of GnRH-II and GnRHR in malignant ovarian tumors as compared with benign ovarian tissues underlies the importance of understanding GnRH-II function in ovarian cancer. In an attempt to define the regulation of GnRH-II in these tissues, we found that a cyclic-AMP responsive element (CRE) is critical for GnRH-II promoter transcription.

Abstract
The present study was conducted to investigate the hypothesis that membrane phosphoinositide breakdown may participate in the actions of luteinizing hormone-releasing hormone (LHRH) on hormone production in the rat ovary. In granulosa cells prelabeled with [³H]-arachidonic acid or [³H]-inositol, treatment with LHRH increased the accumulation of radiolabeled inositol lipids, diacylglycerol and free arachidonic acid, but luteinizing hormone (LH) or cholera toxin did not exert the same effect. Activation of protein kinase C by the phorbol ester, 12-0-tetradecanoyl phorbol-13-acetate (TPA) had a stimulatory action on membrane phosphoinositide breakdown.

Ovarian cancer is the sixth most common cancer and the fifth leading cause of cancer-related death among women in developed countries. Superovulation causes detrimental effects, including embryonic loss and implantation failure. This study determined potential roles for insulin-like growth factor (IGF-I) in uterine environment regulation and preimplantation development, in conjunction with the detrimental effects of superovulation in the rat. IGF-I may be beneficial to preimplantation embryonic development. IGF-I stimulated embryonic development and metabolism in vitro and increased the rate of implantation and fetal development when the blastocysts were transferred into a receptive uterus.

The placenta is an important endocrine organ producing large amounts of steroid and protein hormones which are released into the maternal and fetal circulations. Moreover, it has a very high metabolic rate and consumes a significant proportion of the oxygen and glucose delivered to the uterus and its contents. However, while there have been many studies on the effects of hypoxemia on fetal cardiovascular, metabolic and endocrine functions, there are limited data on the effects of reduced oxygenation on placental endocrine activities. In the present study, we examined the effects of acute (2-h) moderate (maternal Pao₂ lowered by 27-35%, n=5) and severe (maternal Pao₂ lowered by 41-58%, n=4) hypoxemia on placental progesterone output into the maternal circulation in 7 chronically instrumented pregnant sheep (125 to 136 d). Hypoxemia was achieved by lowering maternal inspired O₂ concentration and the hypoxemia period was preceded and followed by 2 h pre-hypoxia and recovery periods, respectively. Control experiments (n=4), involving 6 h periods of normoxia were also carried out.

The ADAMTS (A Disintegrin and Metalloproteinase with TromboSpondin Repeats) are a novel family of secreted metalloproteinases. There is increasing evidence that distinct ADAMTS subtypes play key roles in embryonic development, reproduction and cancer. Nineteen ADAMTS subtypes have been identified in humans but most of them have been characterized only at the structural level. ADAMTS-1, -4, -5, -8, -9 and -15 have been subclassified into a subfamily, known as aggrecanases, owing to their ability to cleave the important extracellular matrix components, versican and aggrecan.

In the present study, the effects of clomiphene citrate (CC) on ovulation, ovarian growth and ovarian steroidogenesis were examined. Ovulation in rats in response to PMSG was completely blocked by administration of three daily treatments of 1.0 mg CC/rat, but was restored by administration of hCG as a preovulatory LH surge substitute. When the number of treatment days was reduced to two days, 1.0 mg of CC enhanced ovulation in response to PMSG, whereas treatment for one day with the same dose of CC did not affect ovulation.

Ritodrine is a beta-adrenergic agonist commonly used to inhibit premature labor contractions in women. The primary goal of ritodrine tocolysis is to prolong gestation, however, other indications may include fetal distress. The purpose of the current study was to examine the metabolic and behavioral effects of ritodrine in the fetus, using the chronically instrumented pregnant sheep as an experimental model. Ritodrine was infused continuously into 11 fetal lambs at a rate of 2.6 ug/minute for a period of 8, 12 or 24 hours. Samples were taken simultaneously at predetermined time periods from a fetal femoral artery, umbilical vein, maternal femoral artery and uterine vein for the analysis of fetal and maternal arterial and umbilical and uterine venous blood gases, acid-base status, hematocrit, ritodrine concentration, uterine and umbilical blood flow, and glucose, lactate and oxygen concentrations and fluxes.

The distintegrin-like and metalloproteinase with thrombospondin repeats (ADAMTS) are members of a gene family of secreted, multidomain and multifunctional proteinases that are able to proteolytically degrade a diverse array of cellular, extracellular and extracellular matrix (ECM) substrates. We examined the presence of ADAMTS in first trimester human placenta and only ADAMTS-12 was present in cultures of invasive extravillous cytotrophoblast (EVT) at significantly higher levels than in poorly invasive JEG-3 chorocarcinoma cells. Immunohistochemistry staining of chorionic villi derived from first trimester human placenta demonstrated that ADAMTS-12 was intensively immunolocalized in the cytotrophoblast layer but weakly immunolocalized in the poorly-invasive syncytial trophoblast layer.

In order to characterize and compare the pattern of inhibin production in the follicular phase between patients of the clomiphene citrate/human menopausal gonadotropin (hMG) and Buserelin/hMG protocols in in-vitro fertilization, serum inhibin levels from 20 patients in each protocol were measured. The patients were matched for age and had tubal diseases only. It was hypothesized that the serum inhibin levels would be different in these two groups due to possible local actions of clomiphene citrate and Buserelin (a GnRH agonist) on the ovary.

In a series of four experiments, the morphology of oocytes and embryos retrieved from immature rats, treated with either a low or high dose of PMSG, and mature, cycling rats was compared to determine whether a superovulatory dose of PMSG has an adverse effect on oocyte maturation and subsequent fertilization and embryonic development in immature rats. Morphological criteria for the assessment of intraoviductal oocyte aging were established in the first experiment.

Abstract
Abnormal DNA methylation at imprinted and non-imprinted genes has been associated with spermatogenesis failure. However, little information is available regarding DNA methylation at those genes in men affected by severe male factor infertility. We hypothesized a higher incidence of aberrant DNA methylation would be present in the ejaculate and testicular sperm of men affected by severe male factor infertility compared to that in fertile control men. Furthermore, we hypothesized abnormal DNA methylation would also affect non-imprinted genes in the sperm of men affected by severe oligozoospermia.

Abstract
Sex hormone-binding globulin (SHBG) binds androgens and estrogens with high affinity, and regulates the distribution of these sex steroids in the blood and other biological fluids. Liver is the primary site of SHBG production, but the human SHBG transcription unit responsible for this is also expressed in proximal convoluted tubule (PCT) epithelial cells in a transgenic mouse model. Unlike hepatocytes which actively secrete SHBG and retain little immunoreactive SHBG within their cytoplasm, an incompletely glycosylated SHBG isoform has been found to accumulate inside PCT cells. These cells are androgen target cells, and the presence of SHBG within them accentuates androgen-dependent regulation of gene expression mediated by the androgen receptor (AR).

Abstract
The common epithelial ovarian tumors appear to arise from the ovarian surface epithelium (OSE), which is a simple squamous-to-cuboidal meso^helium covering the ovary. The exact mechanism of ovarian tumorigenesis is not well known even though this disease is the most frequent cause of cancer death in gynecological malignancies. Repeated ovulation contributes to neoplastic transformation of OSE, indicating that the process of healing ruptured OSE may contribute to the disease. Therefore, it has been hypothesized that endocrine and autocrine factors may influence the occurrence of ovarian tumors in women.
