Abstract
The placenta is an important endocrine organ producing large amounts of steroid and protein hormones which are released into the maternal and fetal circulations. Moreover, it has a very high metabolic rate and consumes a significant proportion of the oxygen and glucose delivered to the uterus and its contents. However, while there have been many studies on the effects of hypoxemia on fetal cardiovascular, metabolic and endocrine functions, there are limited data on the effects of reduced oxygenation on placental endocrine activities. In the present study, we examined the effects of acute (2-h) moderate (maternal Pao₂ lowered by 27-35%, n=5) and severe (maternal Pao₂ lowered by 41-58%, n=4) hypoxemia on placental progesterone output into the maternal circulation in 7 chronically instrumented pregnant sheep (125 to 136 d). Hypoxemia was achieved by lowering maternal inspired O₂ concentration and the hypoxemia period was preceded and followed by 2 h pre-hypoxia and recovery periods, respectively. Control experiments (n=4), involving 6 h periods of normoxia were also carried out.

Abstract
The gonadotropin - releasing hormone (GnRH) receptor is a plasma membrane associated receptor and a member of the GTP - binding protein coupled receptor family. The interaction of the ligand, GnRH, and the GnRH receptor is a critical event in the endocrine control of reproduction. This coupling stimulates the synthesis and release of both luteinizing hormone and follicle stimulating hormone from the anterior pituitary. In addition, GnRH - GnRH receptor binding acts locally to regulate human chorionic gonadotropin secretion in the placenta and steroidogenesis in the ovary. The objective of this thesis was to isolate and characterize the gene for the GnRH receptor in human.

Abstract
In vitro fertilization (IVF) has been achieved in several mammalian species, including humans. Using the technique of IVF and embryo transfer, pregnancies can now be established in infertile women and thousands of live births have occurred. However, many of the embryos transferred to the uterus fail to achieve or sustain implantation. The failure of implantation has raised questions about the possible detrimental effect of superovulation on chromosomal complements. In this study, the hypothesis that chromosomal aberrations exist in mouse preimplantation embryos and human unfertilized oocytes after superovulation, was tested. The incidence of aneuploidy in spontaneously ovulated group (5.6%) was similar to that reported by others.

The patients were matched for age and had tubal diseases only. It was hypothesized that the serum inhibin levels would be different in these two groups due to possible local actions of clomiphene citrate and Buserelin (a GnRH agonist) on the ovary.

Calbindin-D9k is a cytosolic calcium binding protein expressed in the mammalian intestine, placenta, and uterus. The protein is probably involved in calcium transport across the intestinal and placental epithelia. The objective of this thesis was to study the structure of the human and porcine calbindin-D9k at the cDNA and genomic level. The cDNAs for the bovine, murine, and ratcalbindin-D9k as well as the gene for rat calbindin-D9k had been cloned previously.

By using modified hybridoma technology, monoclonal antibodies against an ovarian tumor cell line, OC-3-VGH, were generated in Dr. Lee's laboratory. Among these antibodies, RP 215 was shown to react specifically with a tumor-associated antigen, COX-1. On SDS gel, COX-1 has a molecular weight of 60 KD and exists as an aggregate in the natural- state. A highly purified COX-1 was obtained mainly by immunoaffirtity chromatography, with RP 215 as the affinity ligand, from the shed medium of cultured tumor cells.

The sperm antigen (SA-63) which reacts with HS-63 was found to be localized on the sperm acrosome. Following sperm capacitation, this antigen becomes exposed and is shed after the acrosome reaction. SA-63 may be involved in the sperm acrosome reaction during the initial fertilization process.

A delicate balance of steroid and gonadotrophic hormones is essential for intrafollicular oocyte maturation and successful fertilization and embryonic development. Previous studies have demonstrated that a superovulatory dose of pregnant mare serum gonadotrophin (PMSG) has excessive gonadotrophic activity and alters intrafol1icular steroid hormone levels. In a series of four experiments, the morphology of oocytes and embryos retrieved from immature rats, treated with either a low or high dose of PMSG, and mature, cycling rats was compared to determine whether a superovulatory dose of PMSG has an adverse effect on oocyte maturation and subsequent fertilization and embryonic development in immature rats. Morphological criteria for the assessment of intraoviductal oocyte aging were established in the first experiment.

Abstract
Atrial natriuretic peptide (ANP) of atrial myocyte origin, has been shown to play a role in the diuresis, natriuresis, and antagonism of angiotensin and vasopressin. However, it is now apparent that in addition to the production of the peptide in the heart and in its role in fluid and electrolyte homeostasis, it is also produced in the central nervous system participating in the regulation of pituitary hormone secretion. Administration of ANP through both central and peripheral routes has been shown to inhibit secretion of luteinizing hormone (LH) in the gonadectomized rat model.

Abstract
In the present study, the effects of clomiphene citrate (CC) on ovulation, ovarian growth and ovarian steroidogenesis were examined. Ovulation in rats in response to PMSG was completely blocked by administration of three daily treatments of 1.0 mg CC/rat, but was restored by administration of hCG as a preovulatory LH surge substitute. When the number of treatment days was reduced to two days, 1.0 mg of CC enhanced ovulation in response to PMSG, whereas treatment for one day with the same dose of CC did not affect ovulation.
